Switch #:
SWTI000659
Switch type:
Avidity-sensing

Description:
Orexin-A induced phosphorylation of the ITSM and ITIM motifs in Orexin receptor type 1 (HCRTR1) allows binding of Tyrosine-protein phosphatase non-receptor type 11 (PTPN11) via its two SH2 domains. Mutation of either tyrosine in the motifs abolishes binding of Tyrosine-protein phosphatase non-receptor type 11 (PTPN11).

Participants:
(1) Orexin receptor type 1 (HCRTR1)
(2) Tyrosine-protein phosphatase non-receptor type 11 (PTPN11)

Interactions
Interaction #1 HCRTR1 - PTPN11

Interfaces
(1) LIG_TYR_ITSM motif (79TVTNYFIV86) in Orexin receptor type 1 (HCRTR1)
(2) SH2 domain (112-197) in Tyrosine-protein phosphatase non-receptor type 11 (PTPN11)

Interaction Regulation
PTM-dependent Induction (Phosphorylation of Y83 on Orexin receptor type 1 (HCRTR1)) of the Orexin receptor type 1 (HCRTR1) LIG_TYR_ITSM motif - Tyrosine-protein phosphatase non-receptor type 11 (PTPN11) SH2 domain interaction

Interaction #2 HCRTR1 - PTPN11

Interfaces
(3) LIG_TYR_ITIM motif (356IIYNFL361) in Orexin receptor type 1 (HCRTR1)
(4) SH2 domain (112-197) in Tyrosine-protein phosphatase non-receptor type 11 (PTPN11)

Interaction Regulation
PTM-dependent Induction (Phosphorylation of Y358 on Orexin receptor type 1 (HCRTR1)) of the Orexin receptor type 1 (HCRTR1) LIG_TYR_ITIM motif - Tyrosine-protein phosphatase non-receptor type 11 (PTPN11) SH2 domain interaction
