Switch #:
SWTI000569
Switch type:
Binary
Switch subtype:
Pre-translational

Description:
Alternative splicing removes the mitochondrial localisation signal (MLS) motif of Isoform Mitochondrial of Lysine--tRNA ligase (KARS), abrogating binding to Mitochondrial import receptor subunit TOM70 (TOMM70A) and import into mitochondria. Unusually, the first two exons of Lysine--tRNA ligase (KARS) are non-constitutive. The first exon does not contain a localisation signal (resulting in cytosol localisation) whereas the second exon contains an MLS.

Participants:
(1) Isoform Mitochondrial of Lysine--tRNA ligase (KARS)
(2) Mitochondrial import receptor subunit TOM70 (TOMM70A)

Interactions
Interaction #1 KARS - TOMM70A

Interfaces
(1) ELM:TRG_MLS motif (1MLTQAAVRLVRGSLRKTSWAEWGHRELRLGQLAPFTAPHKDKSFSDQRS49) in Isoform Mitochondrial of Lysine--tRNA ligase (KARS)
(2) Tetratricopeptide repeat (114-578) in Mitochondrial import receptor subunit TOM70 (TOMM70A)

Interaction Regulation
Alternative splicing Abrogation of the Isoform Mitochondrial of Lysine--tRNA ligase (KARS) TRG_MLS motif - Mitochondrial import receptor subunit TOM70 (TOMM70A) Tetratricopeptide repeat interaction
